Lipid nanoparticles (LNPs) have transformed the delivery of RNA-based therapeutics, yet a persistent challenge continues to limit their broader potential. Most formulations default to hepatic accumulation, which creates a significant barrier to extrahepatic targeting. Engineering strategies are increasingly addressing this modern "undruggable" problem through multiple complementary approaches, such as passive targeting and active targeting methods.
