The BioPharm Brief: New Approval, Trial Halt, and Updated Vaccine

First, the FDA has approved PharmaEssentia’s Besremi, or ropeginterferon alfa-2b-njft, for adults with essential thrombocythemia, making it the first new FDA-approved treatment for the rare blood cancer in nearly 30 years. The approval covers adults regardless of genotype or disease status and is supported by the Phase 3 SURPASS-ET trial, where 43% of patients achieved a durable response compared with 6% receiving anagrelide. The therapy also reduced thromboembolic events over 12 months.

In oncology, BioNTech is stopping a phase 2 trial of its individualized mRNA cancer vaccine autogene cevumeran in resected colorectal cancer. The decision follows a recommendation from the trial’s independent Data Safety Monitoring Board, which identified a numerical imbalance in overall survival and concluded that continuing the study was unlikely to change the efficacy outcome. BioNTech said no new safety signals were identified. The company’s separate Phase 2 trial of autogene cevumeran in pancreatic cancer continues.

And ahead of the 2026-2027 vaccination season, Novavax’s updated Nuvaxovid has received approval in the US, EU, and Japan for the XFG variant. The protein-based, non-mRNA vaccine uses Novavax’s recombinant nanoparticle technology and Matrix-M adjuvant. The updated formulation is expected to roll out through Sanofi’s and Takeda’s distribution networks as the new vaccination season begins.
