FDA Grants Fast Track Designation to Consano Bio's Platelet-Derived Biologic C-1101 for Chronic Sciatica

The FDA has granted Fast Track designation to C-1101, Consano Bio's investigational platelet-derived multi-protein biologic for chronic, painful lumbosacral radiculopathy, following the drug's recent IND clearance and as the company advances a Phase 1 trial evaluating the epidural injection.

The FDA has granted Fast Track Designation to C-1101, Consano Bio's lead investigational biologic for the treatment of chronic, painful lumbosacral radiculopathy (LSR), commonly known as chronic sciatica.¹ The designation follows the FDA's clearance of the company's Investigational New Drug (IND) application for C-1101 in July 2026.¹ There are currently no FDA-approved pharmaceutical therapies indicated specifically to treat LSR.¹

Why does LSR represent a significant unmet need?

LSR is a clinically significant subset of low back pain and, together with spine-related pain broadly, is considered a leading cause of years lived with disability in the US, consistent with 2021 Global Burden of Disease data showing low back pain as the leading global cause of years lived with disability.¹'² LSR is also described as a major driver of opioid initiation and chronic opioid use, and for many patients it persists for years and can progress to irreversible nerve damage and permanent disability.¹ Beyond its association with opioid use, LSR is strongly associated with depression, anxiety, and rates of suicidal ideation well above those of the general population.¹

How does C-1101 work?

C-1101 is described as the first pharmaceutical therapy of its kind in development: a novel, platelet-derived multi-protein biologic designed to treat LSR by modulating inflammation and stimulating cellular repair at the site of nerve injury.¹ The product contains consistent concentrations of cytokines, growth factors, and matrix proteins intended to trigger the body's natural healing response, delivered via epidural injection to provide supraphysiologic concentrations of these proteins directly to the injury site.¹ This concept builds on earlier academic research into platelet lysate epidural injections for radicular pain.³

What is the status of C-1101's clinical development?

Consano Bio is advancing C-1101 in a multicenter, randomized, controlled, double-blind phase 1 study (C-1101-101, NCT07264270) in symptomatic patients with chronic, painful LSR, evaluated across three sequential, dose-escalation cohorts, with each participant receiving a single epidural injection.¹ The study is currently enrolling in Australia and is extending into the US following the FDA's IND clearance in July 2026.¹ The Fast Track designation was granted based on the unmet need in LSR and the nonclinical data package Consano Bio submitted in support of C-1101's clinical potential; the FDA's Fast Track program is designed to facilitate development and expedite review of therapies addressing serious conditions and significant unmet medical needs.¹
